چکیده

The treatment of gram-positive bacteria and yeasts with tunicamycin induces morphological changes such as elongation, conversion from rod-form to cocci-form and enlargement of microbial cells. Even when bacteria grew normally or to a somewhat lesser extent than controls as determined bymeasurement of optical density, the increase in viable cells was suppressed to a great extent. Such effects of tunicamycin are observed without inhibition of protein, DNAand RNAsynthesis in Bacillus subtilis W23 cells. RNAand DNAdegradation occurred subsequent to the morphological changes of B. subtilis cells, and the former degraded to a larger extent than the latter. No difference was detected between control and tunicamycin-treated cells in acidsolubilization of the cellular macromolecules labeled with radioactive amino acids or glucosamine.

Allicin attenuates tunicamycin-induced cognitive deficits in rats via its synaptic plasticity regulatory activity

Objective(s): To illuminate the functional effects of allicin on rats with cognitive deficits induced by tunicamycin (TM) and the molecular mechanism of this process. Materials and Methods: 200–250 g male SD rats were divided into three groups at random: control group (n=12), TM group (5 μl, 50 μM, i.c.v, n=12), and allicin treatment group (180 mg/kg/d with chow diet, n=12). After 16 weeks of a...

Tunicamycin induces ubiquitination and degradation of apolipoprotein B in HepG2 cells.

Apolipoprotein (apo) B-100 is an essential component of atherogenic plasma lipoproteins. Previous studies have demonstrated that the production of apoB-100 is regulated largely by intracellular degradation at both the co-translational and post-translational levels and that proteasome-mediated and non-proteasome-mediated pathways are involved in this process. ApoB-100 is a glycoprotein. The pres...

Tunicamycin enhances tumor necrosis factor-related apoptosis-inducing ligand-induced apoptosis in human prostate cancer cells.

Death receptor 5 (DR5/TRAIL-R2) is an apoptosis-inducing membrane receptor for tumor necrosis factor-related apoptosis-inducing ligand (TRAIL/Apo2L). In this study, we showed that tunicamycin, a naturally occurring antibiotic, is a potent enhancer of TRAIL-induced apoptosis through up-regulation of DR5 expression. Tunicamycin significantly sensitized PC-3, androgen-independent human prostate ca...

Investigation of the effect of trimethylamine-N-oxide on the proinflammatory cytokine genes expression in U937-derived macrophages

Background and Aim: Atherosclerosis is known as a multifactorial inflammatory disease. Trimethylamine N-oxide (TMAO) as a risk factor, has a potential to trigger or enhance the immune inflammatory reactions in atherosclerosis. Yet, The exact mechanism by which TMAO induces inflammation during atherosclerosis is not well understood. The present study was designed to evaluate the expression of IL...

Tunicamycin potently inhibits tumor necrosis factor-induced hepatocyte apoptosis.

The protein glycosylation inhibitor tunicamycin protected male BALB/c mice from tumor necrosis factor alpha-induced liver failure. Tunicamycin also inhibited tumor necrosis factor-induced cell death in primary hepatocyte cultures with a median inhibitory concentration of 8 nM, but not in the tumor cell line WEHI 164 clone 13. Hepatocyte death in our culture system was characterized by DNA fragm...
